Acropolis Museum is focused on the finds from the Acropolis, while the National Archeological Museum collects major finds from all over Greece. The Acropolis Museum has a unique design and better views of Athens and the Acropolis. The NAM is much bigger and way more interesting. Do both if you can.

You can visit the Museum before or after the Acropolis (do note you'll need a separate ticket). It comes down to personal preference, however, if you only have one day, I'd recommend visiting the Acropolis first to avoid the crowds. Only after, move on to the Museum.

If you have no mobility problems, then the assent may take you around 15 minutes but you will want to stop a couple of times to take photos. Once at the top it isn't as crowded as the middle stone step area, which gets very congested and can slow you down. Take water with you as on a hot day you will need it. Enjoy.

Although there are water fountains in the Acropolis, we highly recommend that you always carry with you a water bottle as it can get very hot during the summer months. Note that you can only take water inside the Acropolis, other types of drinks are not permitted.
